Çipuranın (Sparus aurata) spesifik bozulma organizmalarının tespit edilmesi

Abstract
Bu çalışmanın amacı ağ kafes ve toprak havuzlarda yetiştiriciliği yapılan çipura balıklarının mikroflora profilini karşılaştırmaktır. Bunun için Muğla Milas’da bulunan iki farklı işletmeden, ağ kafes ve toprak havuzda yetiştirilen çipura balıkları strafor kutularda buzlanarak Muğla Sıtkı Koçman Üniversitesi Su Ürünleri Fakültesi işleme laboratuvarına getirilmiştir. Soğuk depoda (+4°C) 16 gün boyunca muhafaza edilmiş ve depolama boyunca aralıklı günlerde (0, 3, 6, 9, 12, 14, 15, 16) her iki grup balıktan da örnekler alınıp mezofilik, psikrotrofik, lipolitik bakteri ve maya küf ekimleri yapılmıştır. Ekimlerde gelişen kolonilerden izolatlar alınarak Gram boyama, okidaz testi ve API kitleri ile tanımla yapılmıştır. Yapılan ekimlerde ağ kafes örneklerindeki mikroorganizma artışının ilk günlerde toprak havuzdan biraz daha fazla olduğu tespit edilmiştir. Elde edilen sonuçlara göre başta steril olan balık etinde depolamanın ilk günlerinde Gram(+) ve Gram(-) bakterilerin mevcut olduğu ancak depolamanın ilerleyen günlerinde Gram(-) oksidaz pozitif bakteri türlerinin diğer bakterilere göre dominant mikroflora haline geldiği tespit edilmiştir. Ağ kafesten temin edilen çipura balıklarında toprak havuzdan daha geniş mikroorganizma yelpazesine sahip olduğu tespit edilmiştir. Çalışma sonuçları API kitleri ile yapılan tanımlama sonuçları ile değerlendirildiğinde her iki yetişme türü için de bozulma mikroflorasının dominant olarak Pseudomnas spp. olduğu tespit edilmiştir. Çalışma sonunda elde edilen verilerle çipura işletmelerinde depolama süresine bağlı olarak hangi tür mikroorganizmalara dikkat edileceği ortaya konulmuştur. Yetiştirme koşullarının balığın dominant bozulma mikroorganizması üzerine fazla bir etkisinin olmadığı tespit edilmiştir. Bundan sonra yapılacak raf ömrü çalışmalarında bu tür bakterilerin inhibe edilebilmesi için yöntemlerin geliştirilmesi üzerine durulabileceği düşünülmektedir.
