Introducción: En la literatura científica ha sido poco explorada la eficacia de la Anfotericina B y la Rifampicina para la leishmaniasis cutánea. Objetivo: Metaanalizar la eficacia de la Anfotericina B y la Rifampicina en el tratamiento de la leishmaniasis cutánea. Métodos: Revisión sistemática de la literatura con metanálisis en 7 bases de datos multidisciplinarias, se aplicó una estrategia de búsqueda y selección en ensayos clínicos controlados que garantizó exhaustividad, sensibilidad y reproducibilidad. Se realizaron metanálisis para riesgos relativos con efectos aleatorios. Se evaluó heterogeneidad con DerSimonian y Laird, sesgo de publicación con Egger y de Begg. Resultados: Se identificaron 9.825 estudios en Anfotericina B y 5.807 en Rifampicina de los cuales 6 cumplían el protocolo, 3 evaluaron Anfotericina B en presentación liposomal en comparación con antimoniales pentavalentes y 3 compararon Rifampicina contra un placebo; la mayoría de Asia. En los 89 pacientes tratados con Anfotericina B se halló una eficacia del 66,3% (IC95%=55,9-76,7), en los 90 tratados con antimoniales 75,6% (IC95%=66,1-85,0) y en 80 tratados con Rifampicina 67,5% (IC95%=56,6-78,4). Conclusión: A pesar de los resultados favorables para ambos medicamentos es necesario desarrollar estudios en cada país para determinar su eficacia en condiciones epidemiológicas, clínicas y parasitológicas específicas, dada la baja calidad metodológica, el reducido número de investigaciones en América y la elevada heterogeneidad de los estudios analizados.
