Resumo: ß-glucanas fúngicas têm apresentado propriedades imunomoduladora e anticâncer O presente estudo avaliou os potenciais mutagênico, genotóxico, protetor e antioxidante de um exopolissacarídeo obtido do fungo Botryosphaeria rhodina MAMB-5, o botriosferana Também foram investigadas as possíveis alterações promovidas por este exopolissacarídeo na expressão gênica e cinética do ciclo celular A mutagenicidade foi avaliada em linhagens de Salmonella typhimurium Os demais ensaios foram realizados em culturas de linfócitos humanos normais e tumorais (células Jurkat) Em todas as condições avaliadas o botriosferana não foi mutagênico, genotóxico e não reduziu espécies reativas geradas por H2O2 Entretanto, protegeu o DNA de linfócitos normais e tumorais contra danos causados pelo metilmetanosulfonato em condição de pós-tratamento e tratamento simultâneo A citometria de fluxo detectou uma população de células Jurkat tetraplóides em fase G1 após tratamento com botriosferana associado à doxorrubicina Este efeito antiproliferativo pode estar associado à modulação negativa da expressão de genes envolvidos no checkpoint em G1 A repressão do gene CCR5 em linfócitos tumorais tratados com o botriosferana sozinho ou combinado com DXR indica uma possível afinidade do exopolissacarídeo com este receptor Portanto, o botriosferana parece estar envolvido no checkpoint da fase G1 do ciclo e possivelmente com a inibição da expressão do gene CCR5
