Histological, immunohistochemical, biomechanical and wettability evaluations of the Leukocyte- and Platelet-Rich Fibrin membranes derived from dog’s blood

Abstract
O objetivo do estudo foi realizar análises histológica, imuno-histoquímica, biomecânica e de molhabilidade de membranas de fibrina rica em plaquetas e leucócitos (L-PRF), obtidas do sangue de 10 cães hígidos da raça labrador retriever. Coletou-se amostra sanguínea da veia jugular externa com uso de tubo à vácuo sem anticoagulante, que foi imediatamente centrifugado no protocolo de 400g por 12 minutos, empregando centrífuga especializada. Removeu-se o coágulo da L-PRF do tubo e com auxílio de espátula foi raspado o coágulo de hemácias resguardando o “buffy coat”. A membrana da L-PRF foi produzida usando caixa de PRF. Na análise histológica as três porções da membrana foram identificadas, sendo a primeira composta principalmente por eritrócitos, com baixa densidade de leucócitos interpostos entre os mesmos; a segunda formada por leucócitos, predominantemente neutrófilos; e a terceira camada, de aspecto acidofílico, composta pela rede de fibrina. Na imuno-histoquímica todas as amostras apresentaram marcação positiva para VEGF e PDGF, tanto no componente celular, como na malha de fibrina, porém com diferentes intensidades. As avaliações biomecânicas e de molhabilidade foram efetuadas em dois momentos. As membranas L-PRF de 30 minutos suportaram o dobro da tensão máxima em tração quando comparadas com as de 3 horas. A molhabilidade das membranas de 30 minutos foi estatisticamente maior que as de 3 horas. Conclui-se que o protocolo de centrifugação empregado permitiu o desenvolvimento da membrana da L-PRF em cães, comprovado pelas análises histológicas e imuno-histoquímicas, mostrando que o tempo pode influenciar na resistência mecânica e de molhabilidade.
