Abstract
Đinh lăng là một loại cây dược liệu được sử dụng nhiều trong y học do có hoạt tính chống oxy hóa, chống viêm và hạ sốt. Hai yếu tố ảnh hưởng chính đến quá trình trích ly là nhiệt độ (30 - 90oC, mỗi mức độ can thiệp cách nhau 10oC) và thời gian trích ly (10 - 60 phút, mỗi mức độ can thiệp cách nhau 10 phút), được bố trí theo phương pháp giai thừa trong thí nghiệm khảo sát. Phương pháp trích ly có hỗ trợ của vi sóng (Microwave-assisted extraction - MAE) được bố trí theo phương pháp giai thừa với hai nhân tố là công suất (70, 210, 350 W), cùng với thời gian trích ly (10 - 30 giây, mỗi mức độ can thiệp cách nhau 5 giây). Quá trình trích ly sử dụng dung môi nước với tỷ lệ nguyên liệu: dung môi 1: 25 (w/v). Hiệu quả trích ly hợp chất phenolic trong lá đinh lăng được thể hiện qua hàm lượng phenolic tổng số (TPC), flavonoid tổng số (TFC) và khả năng loại gốc tự do DPPH (1,1-diphenyl-2-picrylhydrazyl) trong nguyên liệu trích. Kết quả phân tích hồi quy TPC, TFC và khả năng loại gốc tự do DPPH theo nhiệt độ và thời gian trích cùng với kết quả giải phương trình vi phân tìm nghiệm thức tối ưu cho thấy, với nhiệt độ 60,0oC và thời gian 39,7 phút cho TPC tối ưu (29,1 mg GAE/g), nhiệt độ 63,5oC với thời gian 42,8 phút cho TFC tối ưu (15,1 mg QE/g) và nhiệt độ 55,7oC với thời gian 49,2 phút cho khả năng loại gốc tự do tối ưu (69,0%). Phương pháp trích ly có hỗ trợ vi sóng đạt hiệu quả cao hơn. Với công suất 210 W, thời gian 25 giây, TPC, TFC và khả năng loại gốc tự do DPPH lần lượt là 35,1 mg GAE/g, 22,0 mg QE/g và 79,6%.
