Abstract
RÉSUMÉ: Dans le monde, environ 250 000 personnes sont opérées pour un remplacement de valve cardiaque chaque année. Au Canada, il est estimé que d’ici 2040, près de 1.5 millions de personnes âgées de plus de 65 ans présenteront une pathologie cardiaque associée à une défaillance valvulaire. Les remplacements actuels se font avec deux types de valves artificielles : les valves mécaniques et bioprothétiques. Ces valves présentent des limites en termes de biocompatibilité, durabilité et accessibilité. Ces limites ont conduit à l’émergence de valves polymériques, qui peuvent être conçues sur mesure, leur composition étant modulable pour optimiser leur biocompatibilité et leur propriétés mécaniques. Cependant, elles rencontrent aussi des limites concernant leur durabilité à long terme et une biocompatibilité insuffisante favorisant la thrombose. L’objectif principale de ce projet est de développer des hydrogels à base de PVA/dextrane destinés à la fabrication de valves cardiaques moulées qui présentent une biocompatibilité améliorée tout en maintenant les propriétés mécaniques requises pour l’application aux valves cardiaques. ABSTRACT: Around the world, approximately 250,000 people undergo heart valve replacement surgery each year. In Canada, it is estimated that by 2040, nearly 1.5 million people aged over 65 years will have a heart condition associated with valve failure. Current replacements are made with two types of artificial valves: mechanical and bioprosthetic valves. These valves have limitations in terms of biocompatibility, durability, and accessibility. These limits have led to the emergence of polymeric valves, which can be custom-made, their composition being adjustable to optimize their biocompatibility and mechanical properties. However, they also encounter limits regarding their long-term durability and insufficient biocompatibility favoring thrombosis. The main objective of this project is to develop PVA/dextran hydrogels for the manufacture of molded heart valves that exhibit improved biocompatibility while maintaining mechanical properties required for application to heart valves.
