Preclinical Radiobiological and Immunological Characterization of Syngeneic Head/Neck and Prostate Cancer Tumor Models

Abstract
Previous research has shown that radiotherapy (RT) plus the CXCR4 inhibitor plerixaforimproved primary tumor control and reduced metastases compared to RT alone in patient-derived cervical cancer models. I hypothesized that similar effects would be seen in syngeneic, subcutaneous head/neck (MOC1, MOC2) and prostate (TRAMP-C2) cancer mouse models with an intact immune system. MOC1 and MOC2 but not TRAMPC2 tumors exhibited significant upregulation of the CXCL12/CXCR4 pathway between 24 hours and 96 hours after RT. Several ISGs and the immune checkpoints regulators PDL1 and CTLA-4 were significantly upregulated as well. Furthermore, the macrophage marker F4/80 was significantly upregulated in MOC1 tumors 72 hours after RT. Tumor growth delay was improved in MOC1 tumors with the addition of plerixafor but only during the time period when plerixafor was administered. There was no improvement in growth delay in MOC2 tumors and no change in lung metastases in either model.
