Impact of ZnT8 Deficiency on Insulin Synthesis and Secretion in a Beta-Cell Specific Knockout Model

Abstract
Zinc transporter 8 (ZnT8) is abundantly expressed in beta cells, located on insulin secretory granule membranes. ZnT8 manages high zinc concentrations which contribute to insulin synthesis, crystallization, and secretion pathways, however, impacts of altered ZnT8 activity and accompanying changes to zinc concentrations remains poorly understood. To gain insight into ZnT8 function, ZnT8 beta cell specific knockout (ZnT8 BKO) mice were assessed, which showed elevated insulin secretion capacity when treated with the insulin receptor antagonist S961, which translated ex vivo to increased KCl and glucose stimulated insulin secretion (GSIS) of isolated ZnT8 BKO islets. Interestingly, basal mitochondrial function was elevated, and beta cell-to-beta cell synchrony of calcium influx was reduced, but neither were attributed to elevated insulin secretion. Instead, ZnT8 BKO islets did show a reduction to their elevated GSIS when exposed to added zinc, suggesting enhanced secretion may result from removal of negative feedback of zinc on beta cell secretion.
