Development of Calcium Alginate Hydrogels for the Ionically Triggered Release of an Ophthalmic Wetting Agent

Abstract
This thesis investigates the feasibility of using calcium alginate beads for wetting agent delivery by assessing their encapsulation efficiency (EE), payload retention and release profiles. Beads formulated with the addition of 7.5 mM PEG (10 kDa) or PVP (10 kDa) in the gelation bath showed an increase in EE and payload retention. The properties of the viscosity-modifying polymer layers on calcium alginate beads affected the in vitro release of encapsulated model drug Rhodamine B-Dextran (70 kDa) into simulated tear fluid, enabling modulation of release rates. Meanwhile, storage in high viscosity media such as 50% (v/v) glycerol solution allowed for lower passive release and storing in 450 mM calcium chloride solution led to almost a zero-order release. These results indicate that fine-tuning the cross-linked alginate beads’ formulation and storage condition can produce controlled, ionically triggered drug release.
