The vascular tissues in plants form a continuous network of cell files throughout the plant body (Esau, 1965). The mechanisms and regulatory hierarchies that lead to its localized differentiation are just beginning to be understood. Several lines of evidence including experiments of direct auxin application, auxin transport inhibition and mutants with compromised auxin perception, suggest a role of auxin in vascular differentiation. In addition, auxin can induce rapid and specific changes in gene transcription, which are proposed to be mediated by members of two families, AUXIN RESPONSE FACTOR (ARF) and AUX/IAA (Guilfoyle, 1998; rev. in Hagen and Guilfoyle, 2002). Molecular markers were used to show that in young Arabidopsis leaf primordia, molecular auxin response patterns presage sites of procambial differentiation and the activity of the Arabidopsis gene ARF5/MONOPTEROS/MP, required for proper vascular differentiation, is essential for the regulation of rapidly auxin-inducible AUX/IAA genes, as well as auxin inducible vascular regulators. Regulatory properties of both MP/ARF5 and the closely related ARF7/NPH4 gene seem far more related than suggested by their single mutant phenotypes. The two gene products selectively interact with themselves and with each other, are expressed in overlapping domains, and regulate downstream processes in similar, but quantitatively distinguishable ways. Together these results provide evidence for the biological relevance of ARF-ARF interactions in Arabidopsis plants. Tissue specific auxin responses and potential targets of MP are determined. Consistent with the MP expression pattern, MP is shown to be required for the auxin signal transduction in pericycle cells, for the initiation of lateral roots, conversely, MP is not required for auxin mediated gravitropic responses in the outer tissue layers of the root. To increase the resolution in vascular tissue analysis we identified 34 markers specific to vascular cell and tissue types within a new collection of transactivated enhancer trap lines. The cloning of 23 lines, expression pattern and conditional phenotypes are reported, providing a compendium of markers for different tissues and stages of vascular development which can be used to target cell and tissue specific expression of hetereologous genes.
