Carbonic anhydrase (CA; 4.2.1.1) is a zinc metalloenzyme that catalyzes the reversible hydration of CO2 to bicarbonate. Three families of CA, designated alpha, beta, and gamma, have been identified based on sequence comparisons. All three of these families are represented in the Arabidopsis genome, analysis of which has revealed that as many as 14 putative CA isoforms exist, 6 of these are members of the alpha family, 5 of the beta family and 3 of the gamma family. The purpose of this investigation was to initiate a functional genomics approach to the study of Arabidopsis CAs. This was accomplished by the identification of T-DNA insertion mutants in the putative CAs, the initial characterization of one of these mutants, beta-CA2, and the use of bioinformatics tools to predict the sequence conservation, phylogenetic relationship and subcellular locations of the putative Arabidopsis CAs.
