Abstract

This thesis investigated the role of Nbp2p in the regulation of the Cell Wall Integrity (CWI) MAPK pathway in Saccharomyces cerevisiae. Nbp2p acts as an adaptor protein for the Ptc1p phosphatase, using its SH3 domain to mediate interactions with PxxP-containing target proteins. Co-immunoprecipitation assays provided the first evidence of an interaction between Nbp2p and the MAPK kinase kinase of the CWI pathway, Bck1p. A series of mutations to the Nbp2p SH3 domain and the Bck1p recognition sequence showed a correlation between the affinity of the Nbp2p-Bck1p interaction and the activation of the CWI pathway, and hyperactivation of the pathway was deleterious to cell growth under a variety of cell wall stress conditions. A conserved motif within the Nbp2p N-terminal domain was also identified to be essential for Ptc1p binding. This work demonstrates a novel role for Nbp2p in the CWI pathway and suggests a common targeting mechanism for Ptc1p.
