Elucidating the Interactions of the Sec1/Munc18 Protein VPS33B with SNAREs

Abstract
Platelets are anucleate disk-shaped cells that are involved in a variety of physiological processes such as hemostasis, inflammation and wound healing. Platelet function is associated with abundant protein-bearing secretory vesicles referred to as α-granules. Defects in α-granule biogenesis lead to severe bleeding phenotypes, as observed in early lethal hereditary ARC syndrome caused by loss of function of either VPS33B or VPS16B, which together form a stable functional VPS33B/VPS16B protein complex. In this thesis, the SM protein VPS33B has been found to exhibit a dual mode of interacting with its cognate SNARE, STX13. Through co-immunoprecipitation experiments, VPS33B was found to interact with STX13 directly, or with its associated SNARE complex: STX13/VTI1A/STX6/VAMP4. These findings suggest a role for VPS33B/VPS16B in facilitating homotypic fusion between early endosomes, a key step in the formation of α-granules.
