Abstract
Cortical radial precursor cells (RPs), the neural stem cells (NSCs) of the developing brain, give rise to populations of functionally-specialized cortical neurons and glia arranged in different functional areas to build the neocortex. Despite extensive population-level studies of the neocortex, little is known about the single-cell lineages of RPs and the cell types they produce at the clonal level. The process by which RPs undergo lineage progression, giving rise to different neocortical cell populations throughout embryonic cortical development, remains under studied. In this thesis, I implemented the cell barcoding technique, pbCellTag, a piggyBac transposon-based system developed in our lab allowing for analysis of single RP cell lineages. Tracing RP lineage progression clonally reveals the temporal developmental patterns by which the neocortex is built, identifying clones and the cell types present within them. I further validated the robustness of the pbCellTag method using multiaddressable genome-integrative color (MAGIC) markers and showed that pbCellTag faithfully recapitulates RP lineage progression in vivo.
