Discovery and Characterization of LPAR1 Antibodies for Breast Cancer Metastasis Inhibition

Abstract
Breast cancer is the leading cause of cancer death for women worldwide. Due to the heterogeneity of breast cancers, more diverse treatments are needed. Lysophosphatidic acid and one of its receptors, lysophosphatidic acid receptor 1 (LPAR1), have been implicated in mediating cancer metastasis. Here, we discovered synthetic monoclonal antibodies using M13 bacteriophage displaying human Fab domains selected against virus-like particles expressing LPAR1. One antibody, 17479, bound strongly to LPAR1 by flow cytometry and showed the most promising developability profile from non-specificity enzyme-linked immunosorbent assay (ELISA) and size-exclusion chromatography. 17479 showed potential in inhibiting LPAR1 function in vitro when tested in MDA-MB-231 breast cancer cells in a scratch wound assay, where treatment with 17479 showed a decrease in wound-healing capabilities. Further characterization and development of 17479 and other LPAR1 antibodies can contribute greatly to the field of breast cancer treatment due to their potential therapeutic value in inhibiting metastasis.
