Investigating the Genetics of Depression and Anxiety in a Multi Ancestral Cohort of Children and Adolescents with and without Systemic Lupus Erythematosus

Abstract
Individuals with childhood-onset systemic lupus erythematosus (cSLE) have a higher prevalence of depression and anxiety compared to healthy peers. Genome-wide association studies have separately identified >100 genetic risk loci for SLE, depression, and anxiety. Our study aims to test the association between risk loci for 1) SLE (HLA and non-HLA), 2) depression, and 3) anxiety, and depressive and/or anxiety disorders, in multi-ancestral cohorts of children and adolescents with and without SLE. Of 488 individuals with cSLE, 102 (21%) had a depressive and/or anxiety disorder. Of 6,638 individuals without cSLE, 332 (5%) had a depressive and/or anxiety disorder. SLE, depression, and anxiety polygenic risk scores (PRSs) were not significantly associated with depressive and/or anxiety disorders in either cohort. In individuals without cSLE, a sensitivity analysis suggested an association between the depression PRS and depressive and/or anxiety disorders among individuals of female sex, but not male sex.
