Functional Characterization of Phosphodiesterase Regulating Long Non-coding RNAs

Abstract
Hypertension with brachydactyly type E (HTNB) is a rare Mendelian hypertension (HTN) syndrome caused by missense mutations in the phosphodiesterase 3 gene, PDE3A. Upstream of PDE3A is a long non-coding RNA (lncRNA) locus consisting of three lncRNA annotations. These lncRNAs are highly expressed in arterial tissues and harbor several genetic variants associated with blood pressure traits. Upon deletion of the entire lncRNA locus, more than 3200 genes are differentially regulated, including PDE3A and seven additional PDEs. PDEs hydrolyze cyclic-AMP and cyclic-GMP, to regulate essential intracellular signaling pathways. Consequently, the intracellular levels of cyclic-AMP and cyclic-GMP are increased when the locus is absent. This indicates that the lncRNA locus may regulate multiple PDEs, which in turn, may regulate a myriad of cellular processes. Understanding the regulatory role of this locus could provide insight into the regulatory mechanisms of lncRNAs and can uncover a new target for the development of HTN therapeutics.
