Investigating the Role of Mitochondrial Metabolism in Human Kidney Development and Maldevelopment

Abstract
Kidney maldevelopment is a major cause of chronic kidney disease in children. We have limited understanding of the pathogenesis of kidney maldevelopment and there are no specific treatments. Metabolism is important in the pathogenesis of other types of acquired kidney disease, yet little is known about its role in maldevelopment. It is known that as nephron progenitor cells differentiate into mature epithelial nephrons, a metabolic transition from glycolytic to mitochondrial predominance is seen. This has been demonstrated in murine models, and in human tissue at a transcriptomic and metabolomic level. I have leveraged human kidney organoids as a model to add live functional analysis of metabolism in a human developmental context and have demonstrated that at the time points assessed, mitochondrial metabolism predominates. I have also generated a patient-specific kidney organoid model of HNF1β-related kidney maldevelopment in which to interrogate the role of mitochondrial metabolism in the context of kidney maldevelopment.
