Molecular and genetic analysis of breast cancer metastasis in the human bone NOD/SCID mouse model

Abstract
In this study, we create a clinically relevant model using NOD/SCID mice engrafted with human bone fragments in both right and left flanks, and show that the human bone implants are viable and functional for more than 12 weeks. To investigate the growth and metastatic behavior of breast cancer, human primary breast tumor specimens are subsequently transplanted under only the right flanks of the mouse model. Some of the engrafted tumors proliferate extensively with massive neo-vascularization and metastasize to the initially tumor-free left bone grafts, indicating that this mouse model can be used to distinguish primary breast tumor that do or do not proliferate and metastasize to human bone. To identify genes that correlated with tumorigenesis and metastasis to bone, gene expression profiles using whole genome microarrays are generated by analyzing the differential expressions of the primary patient breast tumors versus the metastatic breast tumors developed in our mouse model.
