The Production and Investigation into Properties of Novel Enzymatically Derived Beta-Linked Manno-oligosaccharides as Bioactive Carbohydrates

Abstract
The valorization of agricultural byproducts into functional bioactive carbohydrates offers an alternative to nutritional supplements and therapeutic approaches to improving health. This research explores the development and application of mannan-oligosaccharides (MOS) derived from copra meal—a low-cost residue of coconut processing—through enzymatic hydrolysis, aiming to advance gut health strategies that have the potential to be scalable and environmentally responsible. The study evaluates MOS as a dual-function bioactive agent: one that supports the growth of health-associated commensals (HAC) and another that inhibits pathogen adhesion. We first examine the physicochemical characteristics of MOS fractions, comparing their molecular weight distributions and prebiotic activity across three of the most widely researched and commercially utilized probiotic strains. Both low and high molecular weight MOS fractions enhance the growth of all tested probiotic strains in vitro, performing comparably to FOS and outperforming XOS and inulin. In vivo administration in mice reveals that 3% MOS supplementation modulates gut microbiota composition, increases evenness, and enriches taxa such as Enterococcus and Lachnospiraceae - which have been linked to improved gut barrier function and reduced inflammation. MOS consumption also influences short-chain fatty acid production and lipid metabolism-related pathways in a sex-specific manner suggesting potential implications for cholesterol regulation and cardiovascular health. In a separate application, we assess the anti-adhesive potential of MOS against Escherichia coli strains expressing FimH adhesins – which has never been done before using this type of MOS. Competitive binding assays demonstrate that MOS effectively inhibits bacterial adherence and invasion in epithelial cell models, paralleling the effects of a FimH-deficient mutant—highlighting its role as a potential glycomimetic capable of interfering with mannose-specific adhesin interactions. Corresponding reductions in IL-8 secretion further support the anti-inflammatory potential of MOS. Overall, this body of work establishes MOS as a multifunctional dietary component capable of modulating the microbiome and mitigating pathogen colonization and adds evidence to support its prebiotic activity. By integrating circular bioeconomy principles with targeted bioactivity, the findings present pathway for transforming agricultural residues into next-generation microbiome-based interventions.
