Abstract
Imagine a seaweed bed so dense that it drapes the granite boulders of Nova Scotia's Atlantic shoreline in an unbroken olive-brown blanket that is Ascophyllum nodosum at peak biomass, and within its tissues lies a family of polyphenols found nowhere else in the plant kingdom. This research extracted phlorotannin fractions from A. nodosum harvested along the Nova Scotia coast and evaluated their ability to inhibit mushroom tyrosinase, a screening target for cosmetic skin-brightening agents. Kelp was collected during autumn 2021, freeze-dried, and extracted with 60% aqueous ethanol. Phlorotannins were enriched by adsorption on Amberlite XAD-7HP resin and separated into low- and high-molecular-weight pools by Sephadex LH-20 chromatography. Total phlorotannin content was quantified by the 2,4-dimethoxybenzaldehyde (DMBA) assay, and molecular weight distribution was characterised by MALDI-TOF mass spectrometry. The ethyl acetate fraction showed 67.3% tyrosinase inhibition at 100 ?g/mL, outperforming the butanol and aqueous fractions. MALDI-TOF revealed phlorotannin oligomers ranging from trimers (m/z 375) to dodecamers (m/z 1490), with hexamers and octamers dominating the most active fraction. IC?? for tyrosinase inhibition was 34.8 ?g/mL for the ethyl acetate pool, compared with 8.2 ?g/mL for kojic acid.
