Common rs3808607 Polymorphism in the Cholesterol 7α‐Hydroxylase ( <i>CYP7A1</i> ) Gene Determines the Degree of Responsiveness of Circulating Cholesterol Concentrations to Dairy Consumption

Abstract
We have previously demonstrated associations between the presence of the single nucleotide polymorphism (SNP) rs3808607 (-204 T>G) G allele of the CYP7A1 gene, which encodes the bile acid rate-limiting enzyme cholesterol 7α-hydroxylase, and significant reductions in total and LDL-cholesterol (TC and LDL-C) concentrations in response to plant sterol and barley β-glucan consumptions, separately. Our objective was to validate this genotype x nutrient interaction via a higher cholesterol and saturated fat dairy diet consumption. In a multicentre randomized crossover study, 99 normocholesterolemic adults consumed either 3 servings/day of DAIRY products (1% M.F. milk, 1.5% M.F. yogurt, and 34% M.F. cheese) or dairy-free CONTROL products for 28 days as part of a prudent diet. SNPs were assessed by TaqMan genotyping assay. Serum TC and LDL-C concentrations after DAIRY vs. CONTROL were higher (P<0.05) among carriers of the CYP7A1 rs3808607-G allele (GT: +0.20 and GG: +0.28 mmol/L for TC; GT: +0.19 and GG: +0.12 mmol/L for LDL-C) while no effect of DAIRY was seen among the TT homozygotes (-0.11 and -0.03 mmol/L for TC and LDL-C, respectively). These observations complement our findings from previous studies where carriers of the rs3808607-G allele showed higher sensitivity in responses of classic biomarkers of cardiovascular health to other dietary manipulations.
