Fatty Acid Transport Proteins Translocate To The Muscle Membrane During Exercise Before And Following Training

Previous work demonstrated that the content of plasma membrane fatty acid translocase (FAT/CD36) protein was acutely increased (translocation) during prolonged moderate intensity exercise, during which whole body fat oxidation increased 2-fold in a group of untrained men and women. However, there was no increase in plasma membrane fatty acid binding protein (FABPpm) during the exercise. In addition, 6 weeks of high-intensity interval training (HIIT) increased the plasma membrane content of FABPpm, but not FAT/CD36 in women. PURPOSE. This study examined the response of plasma membrane FAT/CD36 and FABPpm during 2 hr of acute moderate intensity exercise, both prior to, and following 6 weeks of HIIT in male subjects. METHODS. Ten recreationally active men trained 3 days/week for 6 weeks (18 sessions) on a cycle ergometer. Training consisted of 10 × 4 min bouts at ∼90% VO2 peak, separated by 2 min of rest (HIIT). Training power outputs were increased to maintain heart rate at ∼90% of maximum. A VO2 peak test was conducted pre and post training, as well as a 2 h ride at ∼65% of pre-training VO2 peak. Two skeletal muscle biopsies were taken at rest and again following 2 hr of cycling and expired gases were sampled throughout exercise. Giant sarcolemmal (plasma membrane) vesicles were prepared from the muscle biopsies. RESULTS. VO2 peak significantly increased by ∼20% (3.8 ± 2 to 4.6 ± 2 L/min) following training and whole body fat oxidation increased during the 2 hr of exercise (pre-training: 0.4 ± 0.1 to 0.9 ± 0.1 g/min; post-training: 0.6 ± 0.1 to 1.0 ± 0.1 g/min). The acute exercise bout prior to training increased plasma membrane FAT/CD36 (40%, n = 10) and FABPpm (30%, n = 9). Following 6 weeks of HIIT, the transport proteins were again acutely increased by prolonged exercise at the same pre-training intensity (FAT/CD36, 32%, n=6; FABPpm, 25%, n = 6)). CONCLUSIONS. Plasma membrane FAT/CD36 and FABPpm content were increased with acute moderate intensity exercise in men before and after 6 wk of HIIT. We believe the increases in plasma membrane fat transport proteins are important for increasing plasma free fatty acid transport into skeletal muscle and ultimately increasing oxidation during prolonged moderate intensity exercise.
