GENERATION OF TRANSGENIC WHEAT PLANTS RESISTANT TO SALINITY

В суровых климатических условиях произрастания многих продовольственных культур необходимо внедрять сорта нового поколения, сорта, созданные методами биотехнологии. Такие сорта уже возделываются в таких биотехнологических странах, как Австралия, США, Канада и с недавнего времени в Аргентине. В настоящее время доказано, что генетические механизмы солеустойчивости определяются десятками генов и факторами транскрипции, работа которых важна для реакций растений на солевой стресс. В своей работе мы использовали ген транскрипционного фактора osgata риса, эктопическая экспрессия которого инициируется избыточным засолением и дефицитом воды. Мы показали, что ген транскрипционного фактора osgata в трансгенной пшенице обеспечивает улучшенную устойчивость к соленому стрессу и показатели роста. Трансгенные растения пшеницы получали путем биобаллистической генетической трансформации конструкцией, несущей полноразмерную копию гена osgata под злакоспецифичным Act1 промотором и селективный ген bar. ПЦР-анализ трансгенных растений Т0 и Т1 подтвердил интеграцию osgata в геном пшеницы. Стабильную экспрессию osgata в геноме пшеницы дополнительно подтверждали полуколичественным анализом ОТ-ПЦР. По сравнению с нетрансформированными растениями, трансгенные растения, экспрессирующие osgata, продемонстрировали лучшие показатели роста и улучшенную защиту от повреждения клеток при солевом стрессе. Soil salinization is one of the main processes of agricultural land degradation. In the harsh climatic conditions of the growth of many food crops, it is necessary to introduce new generation varieties, varieties created by biotechnology methods. Such varieties are already being cultivated in biotech countries such as Australia, the USA, Canada, and more recently in Argentina. It has now been proven that the genetic mechanisms of salt tolerance are determined by dozens of genes and transcription factors, essential for plant responses to salt stress. In our work, we used the rice osgata transcription factor gene, the ectopic expression initiated by excessive salinity and water deficit. We have shown that the osgata transcription factor gene in transgenic wheat provides improved salt stress tolerance and growth performance. Transgenic wheat plants were obtained by bioballistic genetic transformation with a construct carrying a full-length copy of the osgata gene under the cereal-specific Act1 promoter and a selective bar gene. PCR analysis of transgenic T0 and T1 plants confirmed the integration of osgata into the wheat genome. Stable osgata expression in the wheat genome was additionally confirmed by semi-quantitative RT-PCR analysis. Transgenic plants expressing osgata showed better growth performance and improved protection against cell damage under salt stress compared to nontransformed plants.
