The mechanical properties of elastin, including its elasticity, resilience, and stress relaxation, arise from its unique block copolymer structure, comprising hydrophobic (HD) and cross-linking (CLD) domains. To investigate the role of molecular structures of individual hydrophobic and cross-linking domains in modulating mechanical properties, we performed extensive atomistic molecular dynamics simulations of three peptides: hydrophobic [h = (GVPGV)7], cross-linking (x = A5KAAKYGA) domains, and a hybrid peptide (h-x = GVPGVA5KAAKYGA). The study of the h-x peptide allowed us to examine the interplay between hydrophobic and cross-linking domains and their combined effect on structural and mechanical properties. At equilibrium, all peptides sampled heterogeneous and highly disordered structural ensembles. Hydrophobic peptide h predominantly adopts sparse and transient β-turns, while peptides x and h-x form transient α- and 310-helical structures with moderate propensities. Distinct mechanical properties were uncovered for the single domain h and both cross-linking peptides. Peptide h showed high extensibility, low elastic modulus, and elastic recoil primarily driven by the hydrophobic effect, whereas peptides x and h-x displayed greater stiffness and low extensibility consistent with the structural roles of HDs and CLDs in elastin. Peptide h-x shows a biphasic response to strain, with changes in structural properties similar to those of hydrophobic domain h at lower strains and to those of cross-linking domain x at higher strains underlying the decoupling of mechanical properties of h and x domains. Together, these findings support a simple elastic network model attributing elastin's extensibility and elastic recoil to the properties of HDs, while CLDs contribute to its structural integrity. The single domain h undergoes significant stress relaxation characterized by structural rearrangements toward a more homogeneous and disordered conformational ensemble. This relaxation from its extended state is predominantly driven by conformational entropy. Ultimately, this knowledge will help in understanding elastin's remarkable mechanical properties.
