Abstract T cell receptor (TCR) stimulation and IL-4 receptor signaling mediate STAT-6 activation and differentiation of naïve T cells into T helper type 2 (Th2) cells. However, the relationship between TCR stimulation and IL-4-induced STAT-6 activation is poorly understood. STAT-6 is tyrosine phosphorylated (pSTAT-6) in response to IL-4 as well as IL-13, and it also regulates gene expression of these Th2 cytokines. Our data demonstrate that pSTAT-6 is induced in a biphasic manner in splenocytes harvested from OVA-sensitized BALB/c mice and restimulated in vitro with OVA. pSTAT-6 was induced within 2-4h, lost at 12-24h and re-induced at 48-96h. IL-4, but not IL-13, followed similar biphasic kinetics. Inhibition of either IL-4 or the IL-4 receptor selectively during the first phase had no effect on secondary pSTAT-6 and production of IL-4 and IL-13. In addition, while biphasic pSTAT-6 was induced in splenocytes treated simultaneously with OVA and exogenous IL-4, stimulation of splenocytes with IL-4 alone did not induce secondary IL-4 production and pSTAT-6. These data demonstrate that the presence of IL-4 at early time points is neither required nor sufficient for IL-4 production and pSTAT-6 during the second phase. IL-4 production and pSTAT-6 during the second induction phase, on the other hand, are dependent upon OVA-induced TCR signaling. Studies are underway to define the cellular sources of IL-4 and pSTAT-6 as well as the cells required for each phase of the induction.
