Impact of Bpag1/dystonin crosslinking protein deficiency in cardiac tissue

Abstract
Cytoarchitectural disruptions of cardiac tissue often lead to various forms of cardiomyopathies. In the present investigation, we determine whether a deficiency in bullous pemphigoid antigen1 (Bpag1)/dystonin crosslinking proteins causes cardiac defects in mice. Morphological parameters such as heart weight/body weight ratios, left ventricular wall thickness and chamber area were collected from the hearts of post‐natal day‐14 wild‐type (+/+) and Bpag1/dystonin‐deficient (−/−) littermates belonging to the dt Tg4 transgenic line. No significant differences were observed between +/+ and −/− for these morphological parameters. The expression levels of cardiac hypertrophy markers such as atrial natriuretic factor (ANF), b‐type natriuretic factor (BNF), sarcoplasmic reticulum calcium pump isoform 2A (SERCA2a), were assessed by Real‐time RT‐PCR. Compared to +/+, a 2–3 fold increase in ANF and BNF transcripts as well as a 35% reduction in SERCA2a transcripts were detected from the hearts of −/− mice. This expression profile reveals early molecular signs of cardiac tissue abnormalities in mice lacking Bpag1/dystonin. These −/− mice do not survive beyond two weeks post‐natal. Therefore, changes in morphological parameters may not have yet been evident in these young mice.
