Contribution of Membrane Progesterone Receptors α and β to Ventilatory Chemoreflex in Female Mice.

Abstract
We hypothesized that the membrane progesterone receptors (mPR) α and β which are expressed in the brainstem, modulate ventilatory chemoreflex. Adult female mice were instrumented with an osmotic pump for chronic infusion of small interfering RNA (siRNA) against mPRα or mPRβ in the fourth ventricle. Two weeks later, respiratory recordings were performed using whole body plethysmography under normoxia (30 min) and then in response to hypoxia (12% O 2 ), hypercapnia (5% CO 2 ) or hypoxic‐hypercapnia (5% CO 2 + 12% O 2 ), 10 min each. Compared to control experiments (vehicle solution without siRNA), the deletion of mPRα decreased the ventilatory response to hypercapnia (early phase, at 1 min of exposure). Whereas mPRβ deletion suppressed the ventilatory response during the early phase of exposure to hypoxia and hypoxic‐hypercapnia. In mice treated with the siRNA against mPRβ, the ventiltory responses to hypoxia and hypoxic‐hypercapnia were observed after a delay of 5.5±0.7 and 5.1±0.5 min compared to 0.2±0.1 and 0.7±0.4 min in controls. Specific deletions of mPRα and mPRβ have been verified by immunohistochemistry. We conclude that mPRα and β modulate the ventilatory chemoreflex in adult female mice. Founded by CIHR (MOP‐102715).
