Abstract

In this chapter, the theory of dependence of enzymatic reactions on pH is presented. A general model for the effect of pH on enzyme catalysis is derived from first principles. The effects of pH on catalytic parameters, Vmax and Km, are discussed in light of knowledge of the effects of acids and bases on reaction rates. A new graphical method for the determination of pK values of catalytically relevant residues within an enzyme is developed in this chapter as well, and compared to existing graphical methods.
