Vascular reactivity differs in mice lacking lymphoid cells compared to wild‐type mice in a cremaster muscle preparation.

Abstract
A relationship has been established between lymphoid cells and endometrial artery remodeling. Whether these cells have an effect on arteriolar reactivity of other vascular beds has not been investigated. To address this, we examined vascular responses of first order arterioles to endothelial dependent and independent vasoactive compounds in a mouse cremaster muscle preparation in situ of both C57Bl/6 (WT) and alymphoid (Rag; no T‐, B‐ or NK cells) mice. Acetylcholine (ACh, 10‐7 – 10‐5M), Phenylephrine (PHE, 10‐7 – 10‐5M), Adenosine (Ado, 10‐8 – 10‐6M) and Angiotensin II (AII, 10‐8 – 10‐6M) were topically administered in increasing concentrations at 5 or 10 (AII) minute intervals. Peak dilations caused by ACh and Ado did not significantly differ between WT and Rag mice, however the rate of dilation to 10‐6M ACh of Rag (0.5+/−0.1 um/sec) was significantly faster than that of WT (0.1+/−0.1 um/sec). 10–8M AII elicited an initial dilatory response that was significantly greater in Rag than WT (change from baseline diameter: 14.8+/−5.4 vs 1.2+/−2.1 um), but constrictions induced by 10–7 and 10–6M were not significantly different. Constrictions produced by 10–6 and 10–5M PHE were significantly attenuated in Rag (−6.6+/−0.7, −8.9+/−0.7 um) than those in WT (−17.8+/−3.6, −21.5+/−3.1 um). These results suggest a possible role for lymphoid cells in arteriolar development and/or reactivity. CIHR and NSERC.
