The Regulator of G protein Signaling (RGS) proteins serve as GTPase accelerating proteins. We have identified a novel role for RGS2 in the control of protein synthesis that is independent of its established RGS domain function. The objective of this research was to characterize further this new aspect of RGS2 signaling, with the promise of identifying a novel therapeutic target for treating diseases characterized by impaired protein synthesis. RGS2 was found to bind to the eukaryotic initiation factor 2 ε‐subunit (eIF2Bε) in a yeast two‐hybrid screen, and this interaction was confirmed at the protein level by co‐immunoprecipitation. RGS2 was able to inhibit de novo protein synthesis in an in vitro translation assay, whereas RGS1 and RGS4 did not interact with eIF2Bε or have an effect on protein synthesis. The ability of RGS2 to interact with eIF2Bε and inhibit protein synthesis was dependent on a subdomain within its RGS domain, spanning residues 79–116. An RGS2 peptide based on the eIF2Bε‐interacting region was capable of inhibiting protein synthesis in the translation assay.
