Stabilité et compatibilité de l’association ondansétron-méthylprednisolone en solution

Abstract
RESUME Objectif : L’objectif de cette recherche consistait en l’etude de la compatibilite et de la stabilite de l’association ondansetron– methylprednisolone dans une meme poche pour perfusion en vue de la preparation a l’avance de lots de poches, qui pourront etre conservees jusqu’a leur administration aux patients. Methode : Les poches ont ete preparees a des concentrations de 0,16 mg/mL et 2,4 mg/mL d’ondansetron et de methylprednisolone dans deux solvants differents : le glucose a 5 % et le chlorure de sodium a 0,9 %. Deux types de poches (MacoPharma polyolefines et Fresenius Freeflex polypropylene) ont ete evalues pour observer l’impact du materiau sur la stabilite et la compatibilite du melange. Ces poches ont ete conservees a temperature ambiante (22° ± 3 °C, exposees ou non a la lumiere) ou a 4 °C ± 2 °C. La teneur en principe actif a ete determinee par chromatographie liquide haute performance, la compatibilite physique etant evaluee par mirage des poches. Resultats : Notre etude a montre que l’association, dans une meme poche, d’ondansetron et de methylprednisolone, aux concentrations considerees, peut etre conservee pendant 2 jours a temperature ambiante ou 4 jours a 4 °C dans le glucose a 5 % et 3 jours a temperature ambiante ou 4 jours a 4 °C dans le NaCl 0,9 %. La nature du materiau de la poche utilisee (polypropylene ou polyolefines) ainsi que l’exposition a la lumiere n’influencent pas la stabilite. Conclusions : Au vu des resultats obtenus, nous pouvons opter pour une conservation des poches de l’association ondansetron–methylprednisolone au maximum pendant 4 jours a 4 °C (refrigerateur) et 2 jours a temperature ambiante. Ces donnees permettent de preparer les poches a l’avance et de les tenir a la disposition des equipes medicales des la prescription, sans imposer de delai supplementaire lie a la mise en oeuvre de la fabrication.
