Abstract
La tyrosinémie héréditaire de type I est un désordre génétique causé par l'absence d'activité de l'enzyme fumarylacétoacétate hydrolase impliquée dans le catabolisme de la phénylalanine. Le succinylacétone (SA), biomarqueur spécifique, origine de l'accumulation du fumarylacétoacétate causée par la déficience enzymatique. Lors du diagnostic, la concentration du SA chez un patient atteint de tyrosinémie héréditaire de type I varie entre 20 à 700 umol/L et peut facilement être mesuré par des méthodes existantes. Dû à un effet fondateur, l'incidence de la tyrosinémie héréditaire de type I est particulièrement élevée dans la province de Québec. La quantification du SA dans un intervalle de mesure de 1 à 1000 nmol/L dans le liquide amniotique (LA) et dans le plasma ou l'urine des patients traités au NTBC (2-(2-nitro-4-trifloromethylbenzoyl)-1,3-cyclohexanedione) est donc pour nous le but à atteindre. Nous avons atteint ce but grâce à la spectrométrie de masse, en mettant au point une méthode de dosage utilisant un procédé de dilution à l'isotope stable."--résumé abrégé par UMI.
