Glutathione (GSH) Synthesis in healthy Adult Males:Effects of varying Cysteine Intake.

Abstract
Cysteine is generally considered the rate limiting substrate for GSH synthesis. However, there are no studies which have looked at whether GSH synthesis responds to graded levels of cysteine intake. The goal of this study was to determine whether increasing cysteine intake with methionine set at the estimated average requirement (EAR) of 14mg/kg/d and a protein intake of 1g/kg/d will result in increased GSH synthesis. We hypothesized that as cysteine intake increases, GSH synthesis will increase up to a point at which further increases in cysteine will have no effect on GSH synthesis. Fractional (FSR) and absolute synthesis rates (ASR)of GSH were measured in erythrocyte of healthy adult males during an infusion of [ 15 N, 13 C 2 ]glycine. Each subject was studied at 5 levels of cysteine intake. Preliminary results for 2 subjects show that GSH synthesis increases with increasing cysteine intake up to 20mg/kg/d. However, at intakes beyond 20mg/kg GSH synthesis decreased back to baseline levels. Our results suggest that in healthy adults, once the total sulphur amino acid requirements are met for protein synthesis, the body's need for GSH synthesis is also met.
