(1) It was discovered that H9083 has been identified as a tetraploid species of Elymus (StStHH), not a diploid species of Psedoroegneria spicata as stated in the paper referenced above. Genomic DNA from this accession was used as probe DNA, combined with differently labelled genomic DNA from Hordeum (HH) and unlabeled blocking DNA from Elymus (NsNsXmXm), on species of Elymus and Elytrigia. Because of this, only St-genome specific sequences in the StH probe were free to hybridize to chromosomes, the same way as the St probe would have performed. However, the results of the combined probe are still to be interpreted as reported in the aforementioned paper, and results from fluorescence in situ hybridization (FISH) analyses using localized probes have confirmed and redefined the original genomic in situ hybridization (GISH) results.
