Therefore we investigated the functional role of miR-96 in prostate cancer carcinogenesis. METHODS Following transfection with miR-96, LNCaP prostate cancer cells showed a higher proliferation in comparison to control, which could be inhibited by cotransfection with anti-miR-96. Induction of apoptosis with camptothecine showed that the enhanced proliferation could be explained by an inhibited apoptosis in miR-96 overexpressing cells, as measured by Annexin-V FITC/ Propidium Iodide staining. RESULTS In-silico target prediction and Gene Ontology (GO) analysis identified FOXO1 as one possible pro-apoptotic target molecule of miR-96. FOXO1 is a transcription factor, which regulates expression of pro-apoptotic genes, for example Bax and FasL and is controlled by PI3K-Akt kinase signaling. Luciferase reportergene assay proofed miR-96 binding to both predicted bindings sites in the FOXO1 3'-UTR. Overexpression of miR-96 in LNCaP cells resulted in a reduced FOXO1 expression on mRNA and protein level. Quantitative real time PCR and immunhistochemistry of 69 prostate cancer specimens revealed a downregulation of FOXO1 both on mRNA and protein level. CONCLUSIONS This is the first study reporting the anti apoptotic function of miR-96 in prostate cancer. The oncogenic impact of this miRNA can be at least partially explained by its inhibition of FOXO1.
