Abstract
Chronic Obstructive Pulmonary Disease (COPD) is the most common chronic lung disease in North America and is the fourth leading cause of death in the US, Canada and Europe. We believe that the paucity of new mechanistic insights and disease treatments based on this information is due in part to the lack of non‐invasive, sensitive and specific tests for pulmonary structure and function. Our goal is to validate MRI‐derived biomarkers in phantoms and healthy volunteers laying the foundation for the study of lung disease progression in subject volunteers. Our overarching objective is to validate the use of MRI in the detection of early and novel phenotypes and biomarkers of respiratory disease including COPD. Here we report the first results of phantom and human studies with images acquired at 3Tesla. Ten healthy volunteers were scanned under protocol as well as a single pig lung phantom scanned twice within a single week. 3He MRI image‐derived measurements of ventilated volumes of the porcine phantom and subject volunteer images were made three times by a single observer. Using Image J, ventilated volumes were measured (excluding the airways) on the basis of manual outlining of contrast within lung each three times. Four different subject's images were reviewed providing a range of size and age of subjects. One subject was scanned twice on the same day (003) and one was scanned twice within a period of 2 weeks (008). Intra‐observer variability (ICC = 0.99) for all images was assessed.
