Der Verlauf pathophysiologischer Prozesse in Modellen der idiopathischen Lungenfibrose (IPF) ist bislang unzureichend beschrieben. Tiermodelle haben den Mangel, dass zur Bewertung des Fibrosegrades meistens Histologie entnommen werden muss. Ziel dieser Arbeit war es unilaterale bronchoskopische Interventionen durchzuführen, welche das Überleben der Tiere zulassen und anschließed, ähnlich der klinischen Situation, mit Computertomographie fibrotische Prozesse über einen längeren Zeitraum zu beoachten. Sprague Dawley Ratten wurden mit Isofluran anästhesiert und die linke Lunge mittels starrem Bronchoskop intubiert. Zunächst wurde anhand von GFP (green fluorescent protein) gezeigt, dass in konstanter Weise das selbe distale Lungenareal erreicht werden kann. Verabreichung von AdTGFbeta vector in die linke Lunge führte zu lokaler Fibrose, was histologisch und durch Genexpression bestätigt wurde. CT imaging und Dichtebestimmung in der Lunge verifizierte ein klar begrenztes Fibroseareal. Physiologische Messungen zeigten interessanterweise eine generell erhöhte Steifheit der partiell fibrotischen Lunge. Wir verfeinern momentan dieses „survival“ Modell der unilateralen Bronchoskopie in Ratten für den Einsatz in Medikamentenstudien. Die gezielte Applikation von Therapeutika inklusive Bioprodukte und Vergleich zwischen behandelter linker und unbehandelter rechter Lunge am selben Tier sind offensichtliche Vorteile dieser Methodik. Ausserdem kann die Dynamik fibrotischer Prozesse mithilfe von CT und physiologischen Messungen detaillierter charakterisiert werden, um die Lücke zwischen Tiermodell und Klinik weiter zu schließen.
