Allele frequencies/Mendelian inheritance: Allele frequencies of SNP T-233C and SNP C641T were estimated in samples of unrelated (no full sibs) animals from six pig breeds (Table 2).Mendelian inheritances at the two SNP loci were confirmed in a three-generation population of White Duroc • Chinese Erhualian intercross.Association analysis: As the allele C of SNP C641T was fixed in the Duroc population, possible associations of the gene variant T-233C with backfat thickness were only calculated for the individuals of a purebred Duroc population using the General Linear Model procedure of the SAS package (version 8.2; SAS Inc., 2001, Cary, NC, USA).The model used genotype as fixed effect and sex as covariate.Twenty-two homozygotes of genotypes CC, 10 TT-homozygotes and thirtyseven heterozygotes were detected.No significant correlations of the T-233C polymorphism with backfat thickness were observed in the purebred Duroc population (P > 0.05, data not shown). Comments:The physiological role of the SCD gene in the lipid metabolism supports the hypothesis of SCD as a candidate gene for fatness traits in livestock.Nevertheless, the SNP T-233C located in the promoter was shown to have no major effects on backfat thickness in the Duroc population, indicating that the mutation may not alter the SCD expression.Additional polymorphisms of the porcine SCD gene have been recently characterized and deposited in the database (GenBank accession no.AY487830).Further association studies using appropriate populations may be worthwhile.
