Abstract
メイラード反応(グリケーション)はタンパク質と糖など親水性分子同士の研究がほとんどであったが,著者らはこの反応が脂質と糖の間でも起きることを発見した.この発見は,高血糖状態で細胞小器官の膜リン脂質が糖化され,これが糖尿病などの疾病の増悪化に関与するという新しい生体内反応の可能性を示唆した.その後の研究で,糖化脂質が酸化ストレス(脂質過酸化)や血管新生の原因物質であることを突き止めた.更に,高純度な糖化脂質の調製技術と高感度定量法の開発に成功し,糖化脂質が糖尿病者の血しょうに高濃度に蓄積している証拠を見いだした.高血糖に付随する様々な障害が脂質グリケーションを介して進む可能性があり,本稿では糖化脂質の分析技術を述べながら,生体膜脂質グリケーションの実態と,脂質過酸化・細胞機能障害へのかかわりを明確にしようとする著者らのアプローチを紹介する.
