Effect of ETA receptor antagonist on hypertrophy and collagen deposition in heart and aorta in response to chronic aldosterone infusion

Abstract
Hypertension together with increased endothelin-1 or aldosterone (Aldo), may be associated with promotion of cardiac and vascular hypertrophy and collagen deposition. Endothelin antagonism may attenuate collagen deposition and reduce hypertrophy in salt-sensitive hypertension. We evaluated whether the selective ETA receptor-antagonist BMS 182874 (BMS) prevents cardiac and vascular collagen deposition and hypertrophy in aldosterone-infused rats. Rats received subcutaneous Aldo (0.75mg/h) and 1% NaCl in drinking water with or without BMS (40mg/kg in food) for 6 weeks. Heart and aorta were studied histologically after staining with Sirius red. Interstitial and perivascular collagen volumes (IC, PVC) were quantified using videomorphometry, and the left and right ventricular (LV, RV) mass measured and aorta area analyzed. Collagen content: see Table (%). Vascular and LV hypertrophy in experimental rats were significantly reduced by treatment. Thus, ETA receptor antagonist prevented collagen deposition and hypertrophy in LV and RV and aorta in aldosterone-infused rats.
