ITH the object of producing a new cell line for virological purposes W cell cultures were set up from kidney tissue of a female and a male elk.Determination of the chromosome number and constitution of these cells was considered to be interesting from two points of view: It would make it possible to follow any transformation of the cell strains during later passages.Secondly, the karyotype of the elk has not been reported up to now. * CELL CULTURESTwo cell samples were studied: (1) female kidney cells derived from a one-year-old female elk, and (2) male kidney cells from a two-yearold male elk.Cell cultures were set up after trypsinization with 0.25 % trypsin.The growth medium consisted of 20 per cent of calf or elk serum and 80 per cent of Eagle's Minimum Essential Medium (Orion Oy).Good cell growth started soon after explantation in both cell cultures.The growth of the cells was of the epithelial type.To date the female cultures have been passed 17 times, approximately once a week.Preliminary investigations on the virological spectrum of this cell culture are in progress.The chromosomes were studied from the primary culture of the female cells and after the first passage of the male cells.The harvesting procedure was as follows: Colcemid (Ciba) was added to a final concentration of lo-'' three hours before the harvest.The cells were trypsinized with 0.25 % trypsin and hypotonic treatment was achieved by adding three times the volume of distilled water to the trypsin suspension.60 % acetic acid and 0.1 M HC1 fixative were used for 30 minutes.The cells were then suspended in 2 % acetic orcein and preparations were made by manual squashing.
