Avian hepatitis E virus in an outbreak of hepatitis–splenomegaly syndrome and fatty liver haemorrhage syndrome in two flaxseed-fed layer flocks in Ontario

Two commercial layer chicken flocks that were fed a flax-based diet beginning at 28 weeks of age for the production of omega-3 fatty-acid-enriched eggs experienced increased mortality when the birds reached 37 weeks. The average weekly mortality was 0.34% over a 20-week period, with peak mortality of 0.9% for 1 week. Reduced feed consumption, reduced body weight gain and poor peak production were noticed prior to the onset of increased mortality. A total of 245 birds were necropsied and 78% of these had lesions in the liver and spleen, with 44% of those necropsied having changes consistent with hepatitis–splenomegaly syndrome, with lesions ranging from acute periportal lymphoplasmacytic hepatitis to chronic severe cholangiohepatitis with haemorrhage, vasculitis and amyloidosis. A total of 11% of the birds had lesions typical of fatty liver haemorrhagic syndrome, and 22% had lesions found in both hepatitis–splenomegaly syndrome and fatty liver haemorrhagic syndrome. No significant bacteria or viruses were recovered from samples of the liver/bile or spleen but 11 of 21 bile samples contained avian hepatitis E virus RNA detectable with a reverse transcriptase-polymerase chain reaction assay. Comparative sequence analysis found identities of 82 to 92% and 78 to 80% between the helicase and capsid protein genes, respectively, of the virus detected in this outbreak and those of other avian hepatitis E virus isolates, suggesting extensive genetic heterogeneity in avian hepatitis E viruses in Ontario flocks. La mortalité a augmenté quand les animaux atteignaient l'âge de 37 semaines. La mortalité moyenne hebdomadaire a été de 0,34% sur une période de 20 semaines, avec un pic de mortalité de 0,9% durant une semaine. Avant le début de l'augmentation de la mortalité il a été remarqué une diminution de la consommation, une diminution du gain de poids du corps et un pic de production médiocre. Deux cent quarante cinq animaux ont été autopsiés et 78% d'entre eux ont présenté des lésions du foie et de la rate, dont 44% qui avaient des lésions compatibles avec un syndrome de splénomégalie hépatite (HSS), avec des lésions allant d'une hépatite lymphoplasmatique périportale aiguës à une cholangiohépatite chronique grave avec hémorragie, des lésions de vascularite et d'amyloïdose. Onze pourcent de ces animaux avaient des lésions typiques du syndrome foie gras hémorragique (FLHS) et 22% avaient des lésions correspondant à la fois au HSS et au FLHS. Aucun virus ni bactéries importants n'ont été isolés des échantillons de foie/bile ou de rate mais 11 des 21 échantillons de bile contenaient l'ARN du virus de l'hépatite E (HEV) détecté par RT-PCR. L'analyse comparative des séquences des gènes de la protéine de l'hélicase et de la protéine de la capside du virus détecté dans ce cas a montré respectivement 82-92% et 78-80% d'identité avec les autres souches d'HEV aviaires, suggérant une hétérogénéité génétique très grande des HEVs aviaires dans les troupeaux en Ontario. Davon wiesen 78% Leber- und Milzveränderungen auf, die bei 44% denjenigen beim Hepatitis-Splenomegalie-Syndrom (HSS) entsprachen mit Läsionen, die von einer akuten periportalen lymphoplasmatischen Hepatitis bis hin zu einer hochgradigen chronischen Cholangiohepatitis mit Hämorrhagien, Vasculitis und Amyloidose reichten. 11% der Tiere hatten Veränderungen, die typisch für das hämorrhagische Fettlebersyndrom (FLHS) waren und 22% der Legehennen zeigten Veränderungen, die sowohl beim HSS als auch beim FLHS zu finden sind. In den Leber- und Galleproben konnten keine pathogenen Bakterien oder Viren isoliert werden, aber mittels Reverse Transkriptase-Polymerasekettenreaktion wurde in 11 von 21 Galleproben RNS des aviären Hepatitis E-Virus (HEV) nachgewiesen. Vergleichende Sequenzanalysen ließen eine 82–92%ige Übereinstimmung zwischen der Helicase und eine 78–80%ige Übereinstimmung zwischen den Kapsidproteingenen des bei diesem Ausbruch entdeckten Virus und denen von anderen aviären HEV-Isolaten erkennen. Dies läßt auf eine erhebliche genetische Heterogenität bei den aviären HEVs in Hühnerbeständen in Ontario schließen. Se necropsiaron un total de 245 aves y el 78% de éstas tenían lesiones en el hígado y bazo, y un 44% de las necropsiadas tenía cambios compatibles con el síndrome de hepatitis-esplenomegalia (HSS), con lesiones que variaban entre la hepatitis linfoplasmocitaria periportal a la colangiohepatitis crónica grave con hemorragia, vasculitis y amiloidosis. Un 11% de las aves tenía lesiones características del síndrome hemorrágico del hígado graso (FLHS) y un 22% tenía lesiones que se han encontrado en ambos HSS y FLHS. No se aislaron bacterias ni virus significativos de las muestras de hígado/bilis o bazo, pero 11 de las 21 muestras de bilis contenían RNA del virus de la hepatitis B aviar detectado mediante la prueba de la reacción en cadena de la polimerasa-transcriptasa. Los análisis comparativos de secuencias encontraron similitudes del 82-92% y del 78-80% entre los genes de la helicasa y de la proteína de la cápside, respectivamente, de los virus detectados en este brote y los de otros aislamientos de HEV aviares, lo cual sugiere una heterogeneidad genética extensa en los HEV de los lotes de Ontario.
