Makanan merupakan suatu hal yang sangat penting bagi kehidupan manusia. Keberadaan bakteri Escherichia coli dalam sumber air atau makanan merupakan indikasi pasti terjadinya kontaminasi. Tujuan dari penelitian ini adalah mengetahui hubungan kondisi higiene dan sanitasi dengan jumlah kuman dan kandungan E. coli pada nasi bungkus. Jenis penelitian adalah explanatory research dengan metode observasi dan pendekatan Cross Sectional. Sampel dalam penelitian ini adalah total populasi yaitu semua pengolah nasi kucing berjumlah 14 pengolah. Uji statistik yang dipakai adalah Uji Chi-Square. Berdasarkan penelitian ini dapat diketahui sebagian besar pengolah makanan memiliki praktik pengolahan baik, sedangkan hasil analisis diketahui tidak ada hubungan antara praktik pengolahan makanan dengan keberadaan E.coli (p= 0,929). Dari penelitian ini dapat disimpulkan sebagian besar pengolah makanan memiliki sanitasi pengolahan baik, sedangkan hasil analisis diketahui tidak ada hubungan antara sanitasi alat (p=,0,857), air pencucian (p= 0,929) dan tempat (p=0,5) dengan keberadaan E.coli.
