R-GECO1 is a red-shifted calcium indicator (Zhao et al. 2011). Besides being suitable for monitoring the activity of identified GFP-labeled neurons, it could also be useful for optogenetic neural circuit mapping where Chr2 expressing neurons can be activated using blue-light. To apply this new tool in <em>Drosophila</em> neurobiology we have made a UAS-R-GECO1 transgenic stock so that R-GECO1 can be expressed in many different parts of its nervous system using the Gal4/UAS system (Brand and Perrimon 1993). <strong>A.</strong> The coding sequence of R-GECO1 was first PCR amplified from the CMV-R-GECO1 plasmid (donated by Robert Campbell, Univ. of Alberta, Edmonton, Canada) using the following oligos:1.FWD: 5'-GAGAGGTACCAATCAAAATGGTCGACTCTTCACGTCGTAAGTG-3' and REV: 5'-GAGAGGATCCTACTTCGCTGTCATCATTTGTACAAAC-3'. This PCR product [KpnI-AATCAAA-R-GECO-BamHI, ~1.2 kb] was digested with KpnI + BamHI restriction enzymes and subcloned into the KpnI &amp; BamHI sites of the pJFRC14-MCS-attB vector (modified from Pfeiffer et al. 2010). This transgene was targeted for insertion into the <em>Drosophila</em> second chromosome (attP40 site) using the PhiC31 integrase (microinjection by BestGene Inc.). <strong>B. </strong>The expression of R-GECO1 in the larval CNS (middle panel). Flies containing the UAS-R-GECO1 transgene were crossed to the Cha-Gal4, UAS-GFP recombinant line. The CNS of progeny larvae were visualized using an epifluoresence microscope with appropriate optical filter settings. Arrows indicate fluorescence in cell bodies.
