この総説では、宿主-寄生生物間における糖鎖の重要性を示す一例として、Trichinella spiralis という寄生虫を扱う。T. spiralisのN-結合型糖鎖に関する研究は、寄生の機構を明らかにするためになされた、構造、合成、免疫学的手法等を用いた学際的アプローチの結集と言える。他の多くの線虫と同様、Trichinella はその表皮上にN-結合型糖鎖を豊富に含む糖タンパク質群を分泌し、提示している。これら糖鎖のうち大部分は線虫、さらには真核生物全体に共通するものだが、最も興味深い糖鎖はTrichinella に独特な構造をもつ。前者は高マンノース型や切り詰め型、後者は複合型N-結合糖鎖に属する。Trichinella を含む多くの寄生虫では、lacdiNAc構造がアンテナ鎖の組み立て部品として好んで用いらる。T. spiralis のL1幼虫では、このlacdiNAc構造がさらにホスホコリンやD-チベロース (3,6-デオキシ-D-arabino-ヘキソース) で修飾されている。両修飾を受けた糖鎖は免疫原性が著しく高い。チベロースは T. spiralis に対する防御免疫の標的となるため、最も掘り下げた研究がなされている。自然界に存在する糖鎖末端の結合様式 (立体化学) を決定するため、α、及びβ結合したチベロースを含む末端四糖の化学合成が試みられた。3,6-ジデオキシ-D-アラビノ-ヘキソースという前例のないキャッピングを導入するには、これまでにない新しい合成法の開発が必要であった。合成糖鎖を用いた結合実験や抗体を用いた実験により、T. spiralis に対する感染防御を果たす抗体の作用機序が明らかにされている。
