Deciphering airway remodeling in asthma: Application of multimodal nonlinear optical microscopy

Abstract
Asthma is characterized as a chronic inflammatory airway disease associated with airway wall remodeling. Airway remodeling involves all tissues of the airway wall and specifically leads to alterations in collagen and elastin morphology and distribution compared non-asthmatic airways. Multimodal nonlinear optical (NLO) microscopy, has shown potential in label-free visualization of key extracellular molecules, such as collagen and elastin. This work details collagen and elastin remodeling in asthmatic airways, and it’spotential to provide information regarding the morphological changes observed in the airways with great resolution and biochemical specificity. Textural features and directional analysis were extracted from the SHG (second harmonic generation) and TPEF (two-photon emission fluorescence) images of histological sections of airways. . The results of this study for the first time show the use of NLO and image directional and texture analysis to understand the 3-dimentional alterations in the tissues elastic and collagen fibres, in asthmatic airways compared to non-asthmatic airways.
