Impact of dietary iron deficiency on tissue levels in developing rats: Differential response to aging, gender and pregnancy

Abstract
The WHO states that iron deficiency (ID) is the preeminent global micronutrient deficiency. In particular, maternal ID is linked to the development of cardiovascular disease in offspring. Despite this, there is limited understanding of the impact of ID with respect to age, gender and pregnancy. METHODS: 7 and 10 wk old ♂ and ♀ Wistar rats were fed control (270ppm) or low iron (3ppm) diet for 10 wks. After 7 weeks, 3 ♀ from each group were bred. Hematocrit (Hct), iron turnover, and tissue levels were measured during and after dietary manipulations and following gestation (24h). Aortic vascular responses (wire myography) in ♂ rats to contractile agonists and vasodilators were assessed. RESULTS: Hct (♂: 47%→31% vs. 34% ♀: 47%→37% vs. 39%) decreased more in the 7–17wk than in the 10–20wk group, respectively. Further, ♂ rats were more susceptible than ♀ at both ages despite significant liver iron depletion in both sexes (♂: 102 ± 18 ppm →11 ± 1 ppm, ♀: 178 ± 25 ppm→21 ± 7 ppm). ID increased aortic sensitivity to phenylephrine and decreased sensitivity to acetylcholine and a NO donor in the 7–17wk, but not 10–20 wk group. CONCLUSIONS: Despite the minimal age difference, the younger animals were more susceptible to ID. This vulnerability suggests that age‐dependent factors must be carefully considered in characterizing the impact of dietary ID.
