Abstract
Eleven main maize cultivars in northeast district of China were sele cted and the immature embryos was used as explants in order to induce callus. Th e effect of four factors, 2, 4-D, BA , L-Pro and CH and their different combinat ion of concentration on type II callus induction were analyzed. The results show ed that optimal medium for type II callus induction was MS adding 4 mg·L-1 2, 4-D, 5 mg·L-1 BA, 750 mg·L-1 L-Pro and 250 mg·L-1 CH, and the highest inductio n rate is 49.66%. 2, 4-D is the key factor of type II callus induction. Differen tiation medium for buds and root were also optimized respectively. Differentiati on rate of callus and the average differentiation buds number of each callus of three genotypes maize were discussed. Genotype Tian1 has the highest, differenti ation rate is 93.14 % and the most buds of explant is 1.35%. The optimum transpl antation medium for regeneration seedling is vermiculite soaked with 1/8 MS.
