Objective To observe the effect of B7 1/2(CD80/CD86) costimulatory molecules on Th1/Th2 cytokine expression level in the mice infected with Schistosoma japonicum, and study the role of B7 1/2 in the shifting of Th1/Th2 immune response and granuloma formation.Methods Spleen lymphocyte cultures were prepared from the mice infected with Schistosoma japonicum at 6, 8, 10 and 12 weeks after infection. The cultures were incubated with Anti CD80 mAb and Anti CD86 mAb for 72h. The ELISA kit was used for the quantitative determination of IFN γ and IL 4 in the spleen lymphocyte culture medium.Results Anti CD80 mAb and Anti CD86 mAb could not significantly affect the IFN γ expression ( P 0 01). But they could significantly inhibit the expression of IL 4 ( P 0 01), and the effect of Anti CD86 mAb is stronger than that of Anti CD80 mAb.Conclusion The inhibition of costimulators could modulate Th1/Th2 cytokine expression level, and B7 2 molecule mainly affected Th2 type immune response. These findings suggested that B7 1/2 as a mediator of Th1/Th2 immune response could be used as a novel potential approach to control the egg granuloma formation of Schistosoma japonicum.
